鸡副粘病毒病病毒特性和基因型的研究及防制 被引量:21

摘要 :用 SPF鸡胚分别从不同患病鸡群分离到 4株病毒 ,经病毒形态、结构、理化和生物学特性、血清学等研究 ,本病毒为禽副粘病毒 型 ;根据国际上规定三项判定新城疫毒力的标准 ,4株病毒均具有与新城疫病毒速发型相似的毒力 ,属于强毒力的毒株 ;人工感染 71周龄 SPF鸡均 10 0 %发病致死 ;应用一株鹅副粘病毒制备的单克隆抗体获具有对禽副粘病毒 型共同的单抗和仅对鹅副粘病毒毒株和鸡副粘病毒毒株呈阳性反应 ,而对新城疫 F16 E8、L asota等毒株不发生反应的特异性单克隆抗体 ;应用 RT-PCR技术对禽副粘病毒 型分离株 F基因的部分片段进行扩增、排序 ,并绘制基因进化树作分析 ,结果表明鸡副粘病毒毒株为基因 型。应用 Y98毒株和 Four strains of virus isolates were got from different diseased chicken flocks using SPF embryos.All were identified as avian Paramyxovirus type 1 by morphological examination,biological property research and serological tests.These four isolates all confirmed to be high virulence like velogenic Newcastle disease virus (NDV) based on mean death time(MDT) of chicken embryos,intracerebral pathogenicity index(ICPI) in 1 day old chicken,intravenous pathogenicity index (IVPI)in 6 week old chicken,which were used to determined the virulence of NDV.100% of 60 week old SPF chicken were dead after artificial infection.Diffenent mAbs were developed from a Goose paramyxovirus;mAbs react with all avian paramyxovirus type 1;mAbs react only with Goose paramyxovirus and chicken paramyxovirus isolates but not with NDV strain F 48 and Lasota.Part of the fusion gene of this chicken Paramyxovirus isolate were amplified by RT PCR and sequenced.A phylogenetic tree base on obtained sequences of reference NDV strains was constructed and showed that this isolate was similer to the genotype Ⅶ NDV virus,a novel genotype NDV.The inactived vaccine made from isolate Y 98 and isolate T 99 could give good protection against infection.
