摘要
目的:建立氧化石墨(Graphene Oxide,GO)纳米粒中多柔比星(Doxornbicin,DOX)含量测定的高效液相色谱方法。方法:采用DiamonsilTM ODS-C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为十二烷基硫酸钠(SDS)溶液(2.88 g·L^(-1))-乙腈(1:1),加入0.5 ml·L^(-1)三乙胺和4 ml·L^(-1)磷酸;流速1.0ml·min^(-1);检测波长254nm。结果:制剂中辅料对DOX含量的测定无干扰,DOX的质量浓度在1~100μg·ml^(-1)。内线性关系良好(r=0.999 8),平均加样回收率为100.03%(RSD=1.5%,n=9)。GO纳米粒中DOX载药量高达2.35 mg·mg^(-1)。结论:含量测定方法准确可靠、简单易行,可用于氧化石墨纳米粒中多柔比星的含量测定。
Objective:To establish an HPLC method for the determination of doxorubicin(DOX)-loaded graphite oxide nanoparticles. Method:A DiamonsilTM ODS-C_(18) column(250 mm×4.6 mm,5μm) was used with aqueous SDS(2.88 g·L^(-1) )-acetonitrile (1:1) including 0.5 ml·L~triethylamine and 4 ml·L~phosphoric acid as the mobile phase at the flow rate of 1.0 ml·min^(-1). DOX was detected at 254 nm.Result:The linear range of DOX was 1-100μg·ml^(-1)(r =0.999 8).The average recovery was 100.03%(RSD = 1.5%,n =9).The DOX loaded in the graphite oxide nanoparticles was as high as 2.35 mg ·mg^(-1).Conclusion: The established method is accurate and simple in the content determination of DOX in graphite oxide nanoparticles.
