摘要
目的 :测量卵巢癌SKOV3 亲本和耐药细胞内阿霉素聚集量。方法 :采用一种简单敏感的改良荧光分光光谱法。结果 :虽然耐药细胞株SKOV3/ADM内阿霉素聚集量经维拉帕米作用后能显著增加 ,但仍然低于亲本细胞SKOV3。结论 :本法快速、方便 ,在肿瘤细胞多药耐药机理及逆转剂研究中具有实用价值。
Objective:[WT5”BZ]To determine intracellular accumulative quantity of Adraimycin(ADM) in SKOV 3and SKOV 3/ADM cells.[WT5”HZ]Method:[WT5”BZ]A simple and sensitive spectrofluorimetric method was used.[WT5”HZ]Results:[WT5”BZ]The intracellular concentration of ADM in SKOV 3/ADM subline cells was significantly higher after co-incubation with Verapamil,which was lower than that of parent SKOV 3cells.[WT5”HZ]Conclusion:[WT5”BZ]This method is fast and simple.It is of practical value in the research of the Mechanism of multidrug-resistance in human cancer cells and the research of reverse agents.
