摘要
目的 :用改良高效液相色谱法测定人血浆中阿昔洛韦浓度。方法 :以甲醇 重蒸馏水 (5∶95 )为流动相 ,检测波长 2 5 0nm ,鸟苷作内标 ,血浆样品采用 2 0 %高氯酸沉淀蛋白后进样。结果 :阿昔洛韦在 0 .0 31~ 2 .0mg·L-1范围内线性关系良好 ,最低检测浓度为 0 .0 1mg·L-1,高、中、低浓度日内RSD分别为 2 .5 % ,1.2 %和 2 .3% ;日间RSD分别为2 .4% ,5 .8%和 2 .7%。结论 :该法操作简便 ,灵敏度高 ,色谱峰分离好。
OBJECTIVE: A simple, sensitive and rapid HPLC method has been developed for the determination of acyclovir in plasma. METHODS: A Waters HPLC instrument with YMC Pack ODS (250 mm×4.6 mm, 5 μm) as column packing was used . The mobile phase was a mixture of methanol and water (5∶95 V/V) with a flow rate of 1.0 ml·min -1 , and the ultraviolet wavelength was set at 250 nm. Guanosine was used as internal standard, 20 % perchloric acid was used to precipitate plasma protein. RESULTS:The calibration curve was linear in the range of 0.031~2.0 mg·L -1 , and the limit of quantification was 0.01 mg·L -1 , RSD was less than 6.0 %.CONCLUSION: This method is simple, rapid and sensitive. It has been used in pharmacokinetics and bioavailability study of acyclovir.
