Beclin1基因增强三阴性乳腺癌细胞BT-549对多柔比星的敏感性 被引量:3

摘要 目的:探讨Beclin1基因对三阴性乳腺癌细胞BT-549增殖及多柔比星作用于癌细胞的影响。方法:脂质体法分别将质粒pDsRed-C1、pDsRed-C1-Beclin1转染于人三阴性乳腺癌BT-549细胞株。转染后一组用多柔比星处理,另一组不做处理。MTT法分析Beclin1过表达对BT-549细胞增殖的影响,流式细胞仪检测细胞凋亡情况。结果:多柔比星处理前后,Beclin1组细胞增殖均慢于阴性对照组和空白组细胞(各P<0.01);细胞凋亡的变化:多柔比星处理前转染pDsRed-C1-Beclin1的BT-549细胞凋亡率为(8.81±1.45)%,高于转染pDsRed-C1细胞组和空白组(P<0.05),多柔比星处理后,Beclin1组细胞的凋亡率为(14.3±1.09)%,亦高于阴性对照组和空白组(P=0.00)。结论:Beclin1基因过表达抑制BT-549细胞的增殖,增强癌细胞对多柔比星的敏感性。
