摘要
南方菜豆花叶病毒(Southern bean mosaic virus,SBMV)是一种既能通过种子远距离传播,又有介体带毒扩散力的危险性病毒。目前,国内尚未发现.SBMV主要危害菜豆、豇豆等在我国种植面积大、范围广的豆科植物。随着进出口贸易和种质交换日益增多,此病毒传入机会大大增加。为了提高检疫技术水平,了解病毒特性和建立快速检测技术,本文对SBMV单克隆抗体进行了初步研究。
Seven hybridoma cell lines secreted IgG1 McAb against Southern Bean Mosaic Virus ( SBMV ) was prepared by routine techniques. The ELISA titers of cell culture supernatant and the ascite fluid were 640-1600 and 105 -106 respectively. The titer of the polyclonal antiserum from either rabbit or mice ascitic fluid was 1024-2048 using agar immunodiffusion test.Viral antigen treated either with SDS of with formic acid did not react with the McAbs. In contrast, viral antigens treated with heat or freezing and thawing gave stronger reaction than the untreated control. In double antibodies sandwitch test ( HDAS-ELISA ) the use of McAb as the first antibody gave the best results. In immunoadsorption electron microscopy experiments,a large number of viral particles could be trapped by the McAbs diluted 1 : 100-1 : 1000 from diseased crude sap stored for two years at 4℃.
