脑活素对周围神经损伤修复的实验研究 被引量:3

摘要 目的研究脑活素对周围神经损伤的修复作用。方法选取50只成年雌性Wistar大鼠,采取钳夹坐骨神经的方法制造周围神经损伤模型,随机分为5组,每组10只,分别为生理盐水空白对照组(A组)、甲钴胺阳性药物对照组(B组)、脑活素低剂量组(C组)、脑活素中剂量组(D组)及脑活素高剂量组(E组)。各组进行相应药物处理一个疗程(10d)后,对各组大鼠进行坐骨神经指数、斜坡实验及小腿三头肌湿重改变等指标评估。制备大鼠坐骨神经半薄切片,行甲苯胺蓝染色,利用显微镜对神经有髓纤维进行计数。结果 D、E组与A组的比较,坐骨神经指数差异具有显著性(分别为P<0.05、P<0.01)。E组与B、C组用药各组间坐骨神经指数的比较差异具有显著性(P<0.05),余未见显著性差异。斜坡实验A组与C、D、E组各组差异有显著性(P<0.05、P<0.05、P<0.01)。在各用药组间对比中,E组较B组差异具有显著性(P<0.05)。大鼠小腿三头肌湿重改变除E组与A组具有差异改变(P<0.05)外,余无统计学意义。镜下观察,B、C、D、E组均与A组有显著差异(P值均<0.01),其中效果最好的E组,与B组对比结果均具有差异性,尤其在平均直径和面积具有显著差异(P<0.01)。结论脑活素对周围神经损伤有修复作用,其中高剂量组具有较好的疗效。 Objective To investigate the effect of cerebrolysin on peripheral nerve regeneration. Methods 50 fe- male Wistar rats peripheral nerve injury models were made through by clamping sciatic nerve. They were randomly divided into five groups( n = 10 ) :saline group( A), mecobalamin group (B), low dose cerebrolysin group (C), moderate dose cere- brolysin group(D) , and high dose cerebrolysin group (E). Experimental drugs were given to each group once a day, sepa- rately. After 10 days ,the following items as sciatic nerve function index (SFI) , slope experiment and tricipital muscle wet weight were detected. The sciatic nerve was prepared to be semithin section, stained by toluidine blue and assessed under microscope. Result The SFI in rats of test group D, E after 10 days more apparently increased more apparently as com- pared with those of the rats of group A ( P 〈 0.05, P 〈 0.01, respectively) after 10 days, while that of group E was better than those of group B, C ( P 〈 0.05 ). In slope experiment, group A had a worse performance than group C, D, E ( P 〈 0.05, P 〈0.05,P 〈0.01, separately). Comparing by group B, group E did better(P 〈0.05). There were no significant differ- ences between groups except for group E and A, the former was better(P 〈 0.05) in the field of tricipital muscle wet weight change. Assessing under microscope, the axon and myelin of the regenerative sciatic nerve in rats of the last four groups are much denser and thicker than those in saline group, and group E ranked first, which had significance variations in compari- son with group B, especially in mean diameter and area(P 〈 0.01 ). Conclusion Cerebrolysin has the effect of promoting regeneration of injured peripheral nerve. The higher the dose, the better the resuhregenerate function is dose dependent.
