胰腺癌组织HIF-1α、Endostatin和VEGF表达及意义 被引量:3

EXPRESSION AND CLINICAL SIGNIFICANCE OF HYPOXIA-INDUCIBLE FACTOR 1,Endostatin AND VASCULAR ENDOTHELIAL GROWTH FACTOR IN PANCREATIC CARCINOMA

摘要 目的了解低氧诱导因子-α(HIF-1α)、内皮抑素(Endostatin)和血管内皮生长因子(VEGF)在胰腺癌组织中表达的改变及其与胰腺癌临床病理特征的关系。方法应用SABC免疫组化方法,检测8例正常胰腺组织和35例胰腺癌组织标本中HIF-1α、Endostatin和VEGF的表达,并分析其与胰腺癌临床病理因素的关系。结果正常胰腺组织中HIF-1α和VEGF不表达,Endostatin阳性表达率为37.50%,胰腺癌组织HIF-1α、Endostatin和VEGF阳性表达率分别为71.43%、77.14%和68.57%,两组比较差异有显著性(χ2=2.415~13.651,P<0.05)。胰腺癌组织HIF-1α、Endostatin表达与肿瘤TNM分期、淋巴结转移有关(χ2=4.418~6.417,P<0.05);VEGF表达与肿瘤大小、TNM分期及淋巴结转移有关(χ2=6.417~8.670,P<0.05)。胰腺癌组织VEGF、Endostatin表达与HIF-1α表达呈正相关(r=0.662、0.409,P<0.05),Endostatin与VEGF表达无相关性(r=0.218,P>0.05)。结论 H IF-1α、Endostatin和VEGF在胰腺癌组织中高表达并与胰腺癌的临床病理特征相关,HIF-1α与VEGF的协同作用可能参与了胰腺癌的发生发展及淋巴结转移。 Objective To study the changes of expression of HIF-la, Endostatin and VEGF in normal pancreatic tissue and their association with clinical features of cancer of pancreas. Methods By using SABC immunobistochemical method, the expressions of HIF-1α, Endostatin and VEGF in eight cases of normal pancreatic tissue and 35 of cancer were detected and the cor- relation among the expressions and clinicopathology of the cancer analyzed. Results No expressions of HIF-1α and VEGF were found in normal pancreatic tissue, and the positive expression of gndostatin was 37.50% ; in the cancer tissue, the expression of HIF-lu, Endostatin and VEGF was 71.43%, 77. 14%and 68. 57%, respectively, the differences between the two groups were significant (x2= 2. 415-13. 651, P〈0.05). The expressions of HIF 1 α and Endostatin were related with TNM staging and lymph node metastasis (x^2=.418-6.417,P^0.05); that of VEGF was associated with tumor size, TNM stage and lymph node me tastasis (x^2 =8. 670,7. 352,6. 417 ;P%0.05). In pancreatic cancer tissues, the expressions of VEGF and Endostatin were positive ly correlated with HIF la (r=0. 662 ,P〈0. 05), no correlation was noted between the expressions of Endostatin and VEGF (r= 0. 218,P〉0.05). Conclusion High expressions of HIF-lu, Endostatin and VEGF in pancreatic cancer tissue are correlated with clinical features of the disease. The joint action of HIF 1α and VEGF may involve in the occurrence, development and lymph node metastasis of this malignancy.
