G_0/G_1期异时相Cyclin B1的表达对细胞凋亡的影响 被引量:4

摘要 目的研究G0/G1期异时相细胞周期素B1(Cyclin B1)的大量表达对细胞凋亡的影响。方法首先构建了四环素可诱导表达Cyclin B1的单克隆细胞株,其次,筛选出能最大程度阻滞细胞于G0/G1期的胸腺嘧啶核苷(Thymidine)浓度,最后,在阻滞的时间内加入四环素诱导Cyclin B1表达,并用免疫印迹和流式细胞仪检测证实后,用膜联蛋白/碘化丙啶(AnnexinⅤ/PI)双染法和API法检测细胞凋亡。结果终浓度为5mmol/L的胸腺嘧啶核苷能阻滞单克隆细胞株在G0/G1期,并至少维持48h;在细胞同步化期间加四环素诱导后,Cyclin B1表达增加,并且G0/G1期特异性细胞凋亡增加。结论 G0/G1期异时相Cyclin B1的表达能诱导细胞凋亡。 Objective To study the effect of unscheduled expression of Cyclin B1 in G0/G1 phase on cell apoptosis. Methods A monoclone cell line expressing Cyclin B1 was constructed by using tetracycline. The concentration of thymidine which could cause Go/G1 phase cell arrest was determined. Tetracycline was used to induce Cyclin B1 expression in the arrested ceils, and cell apoptosis was detected. Results Thymidine (5 mmol/L) could cause and maintain G0/G1 phase cell arrest for 48 h. The apoptosis of arrested cells was significantly increased after tetracycline treatment. Conclusion Unscheduled Cyclin B1 expression in G0/G1 phase may induce cell apoptosis.
