摘要
为探索豆科植物豌豆作为生物反应器瞬时表达外源重组蛋白的可行性,选择豌豆作为宿主植物,构建了含有酸性成纤维细胞生长因子基因的植物瞬时表达载体,以绿色荧光蛋白(GFP)为报告基因,并以叶片注射法进行接种,在豌豆植株中瞬时表达了人酸性成纤维细胞生长因子,接种后在紫外灯下跟踪观察绿色荧光蛋白的表达情况。研究结果表明,接种后8~10d在紫外灯下可观察到GFP的表达,12~14dGFP表达量最大。蛋白质杂交检测结果表明,酸性成纤维细胞生长因子在豌豆植物中成功表达,说明豌豆植物可作为植物瞬时表达外源蛋白新的选择平台。
In this research, pea plants were used as bioreactor to express acidic fibroblast growth factor (aFGF), and the vector contain acidic fibroblast growth factor (aFGF) was constructed and introduced into pea plants by leave injection; GFP was used as report gene in this system and western blot was used to analyze the expression of foreign protein. The results showed that GFP was visible in plants at 8-10 days post infiltration (dpi.), and the highest accumulation of green fluorescence appeared at 12-14 dpi., Western blot indicated that the aFGF expressed successfully in pea plants.
