叉头框转录因子O亚族1基因对猪肾周脂肪沉积代谢的影响 被引量:7

Effect of Forkhead Box Transcription Factor O1 Gene on the Perirenal Fat Metabolism of the Pigs

摘要 叉头框转录因子O亚族1(forkhead box transcription factor O1,FoxO1)在细胞增殖、分化过程中发挥着重要的作用。采用实时荧光定量PCR技术,对不同月龄民猪和长白猪肾周脂肪中FoxO1、过氧化物酶体增殖物激活受体(peroxisomeproliferator-activated receptorγ,PPARγ)和CAAT区/增强子结合蛋白α(CAAT/enhancer binding proteinα,C/EBPα)基因mRNA表达量进行检测,探讨FoxO1在猪肾周脂肪代谢过程中的作用。结果显示,2品种猪肾周脂肪2.5月龄FoxO1基因mRNA表达量极显著高于1、6、8月龄(P<0.01);分析FoxO1与PPARγ、C/EBPα基因的表达关系,结果发现在2.5月龄后,FoxO1mRNA的表达与PPARγ、C/EBPα相反,说明FoxO1可能通过抑制这2个基因的表达来减少脂肪沉积。 Forkhead box transcription factor O1 (FoxO1) played a critical role in cell proliferation and differentiation of mammals. An experiment was conducted to determine the mRNA expression of FoxO1 ,PPAR 7 and C/EBPa from perirenal fat of pigs by using Real-time PCR to study the role of FoxO1 in fat metabolism. The results indicated that in perirenal adi- pose,the expression of FoxO1 mRNA in Min pigs and Landrace of 2.5 months of age was significant higher than that in pigs of 1,6 and 8 months of age (P〈0.01). According to the analysis of the relationship between mRNA expression of FoxO1 and PPAR7, C/EBPa gene, the expression trend of FoxO1 was contrary to PPARγ,C/EBPa after 2 months, which manifested that FoxO1 inhibited fat deposition through inhibiting PPARγ, C/EBPa mRNA expression. The results suggested that FoxO1 played an inhibitting role in fat deposition of pigs.
