摘要
通过运用细胞的分离培养技术、MTT法、ELISA检测和实时荧光定量PCR技术,比较不同剂量黄芪杂多糖(AHPS)注射组与空白对照组的仔猪血液内的T淋巴细胞和单核细胞的增殖活性、IL-6含量;骨髓中抗菌肽PR-39基因的表达水平,研究AHPS对仔猪内源性抗菌肽PR-39的表达调控作用。试验结果表明,AHPS对仔猪血液中的单核细胞和T淋巴细胞有促进作用;IL-6含量在同一时间内呈剂量依赖性增加,随着注射时间的延长逐渐增多;AHPS对PR-39的表达呈显著地上调作用且呈剂量依赖性。根据相关文献和本试验结果推测,黄芪杂多糖可能通过调控IL-6和PR-39的表达提高仔猪的免疫力。
To study the effects of astragalus heteropolysaccharides (AHPS) on the expression of IL-6 and PR-39 of piglets, the proliferation activities of T lymphocytes and monocytes in peripheral blood were determined by MTT, the IL 6 contents in serum were detected by ELISA, the expression levels of PR-39 in marrow were analyzed by RT-qPCR. The results showed that AHPS could promote the proliferation activities of T lymphocytes and monocytes in peripheral blood. IL-6 contents in ser- um showed dose-dependent patterns at the same time, and increased significantly with the extension of the injection time. AHPS could increased the level of PR-39 mRNA and showed the dose-dependent patterns. According to the relevant literature and the results, we inferred that AHPS might improve the immunity of piglets by increasing the content of IL-6 and the expression level of PR-39.
