摘要
目的观察新生SD大鼠心肌细胞内脂素的表达情况,为临床心肌病治疗的新靶点奠定理论基础。方法出生2~3 d的SD乳鼠,提取并培养原代心肌细胞,培养48 h后换为无血清的培养基继续培养24 h,之后应用RT-PCR、Western-Blot技术检测心肌细胞中内脂素表达情况。结果心肌细胞中内脂素mRNA和蛋白均有表达。结论除脂肪组织、活化的淋巴细胞、单核细胞、中性粒细胞、胎膜上皮细胞以及骨骼肌等细胞以外,心肌细胞也能够表达内脂素。
Objective It is to observe the expression of visfatin in myocardial cells of SD neonatal rats, to establish the theoretical foundation for new targets of the treatment of cardiomyopathy. Methods Ventricular cardiomyocytes were dissociated and cultured from the 2 to 3 - day old neonatal SD rats. In forty-eight hours after seeding, the cardiomyocytes were cultured with the serum-free culture medium for another twenty-four hours. After that, visfatin expression in cardiomyocytes were deter- mined by RT - PCR and Western-Blot technology. Results The mRNA and protein expression of visfatin were both detected in cardiomyocytes. Conclusion Except adipose tissue, actived lymphocyte, monokaryon, neutrophil, embryolemma epithelial cell and skeletal muscle cell, cardiomyocytes can also express visfatin.
