摘要
目的 鉴定辽宁、吉林地区恙虫病东方体分离株的基因型。方法 采用 PCR/ RFL P技术对辽宁、吉林地区恙虫病东方体分离株的 sta5 6目的基因进行分析研究 ,并与国际参考株进行比较。结果 辽宁、吉林地区 6株分离株的 PCR特异性产物经 Hinf 、Hha 、Rsa 酶切后呈现 2种不同的 RFL P图谱 :HCAa92 0 2株、HCAp92 0 3株及 HCM95 0 1株与 Gilliam参考株具有相似的酶切图谱 ,但缺乏 Hinf 的酶切位点 ;KDCt940 2株、KDCp940 3株及 KDCt940 4株与 Karp参考株具有相同的酶切图谱。结论 KDCt940 2株、KDCp940 3株及 KDCt940 4株属于 Karp型 ;HCAa92 0 2株、HCAp92 0 3株及 HCM95 0 1株属于 Gilliam型 ,但基因序列可能存在遗传差异。
Aim\ Determined the gene type of isolated strains of Orientiatsutsugamushi(Ot) in Liaoning and jilin areas.Methods\ PCR/RFLP were used to analyse their restriction maps of sta 56 gene of the isolated strains and made a comparison between the isolated strains of Ot and the international refernce strains.Results\ The distinctive PCR products of the 6 isolated strains from Liaoning and Jilin areas through restriction with HinfⅠ,HhaⅠ,RsaⅠappeared two different kinds of RFLP restriction map.The strains HCAa9202,HCAp9203 and HCM9501 had the similar restriction maps with reference strain Gilliam,but they did not have restriction site of HinfⅠ;Strains KDCt9402,KDCp9403 and KDCt9404 had the simular testriction map with reference strain Karp.Conclusion\ Strains KDCt9402,KDCp9403 and KDCt9404 may be classified into the Karp type;Strains HCAa9202,HCAp9203 and HCM9501 may be classified into the Gilliam type.But the isolated strains had some difference in gen sequence compared with the standard strains.
