丁胺卡那霉素对豚鼠耳蜗毛细胞凋亡及听力阈值的影响 被引量:4

摘要 目的 :探讨丁胺卡那霉素作用于耳蜗系统时毛细胞凋亡现象及其在听力损伤中的作用。方法 :15只豚鼠随机分为 3组 ,各组肌内注射丁胺卡那霉素 40 0mg·kg-1·d-1,分别于用药 1d ,3d ,6d后处死。处死前检测其听脑干反应 (auditorybrainstemresponses,ABR)的变化 ,并利用光镜和TUNEL标记技术检测耳蜗毛细胞发生凋亡的情况。结果 :①豚鼠肌内注射丁胺卡那霉素 1d后耳蜗毛细胞即出现凋亡现象 ,连续用药 3~ 6d ,毛细胞凋亡呈强阳性表达 ;②耳蜗毛细胞凋亡出现的早期豚鼠听力阈值无明显改变 ,连续用药 6d后听力阈值则明显提高。结论 :①丁胺卡那霉素作用于耳蜗毛细胞可引起毛细胞凋亡 ,细胞凋亡是豚鼠耳蜗毛细胞损伤的一条途径 ; Aim: To investigate the cochlear hair cell apoptosis induced by the ototoxic aminoglycoside antibiotic amikacin and the effect of hair cell apoptosis on hearing loss in guinea pigs. Methods: Fifteen guinea pigs were randomly divided into 3 groups,which had been intramuscularly injected with amikacin for 1 day,3 days and 6 days respectively before they were put to death. Auditory brainstem response (ABR) and the apoptosis of cochlea hair cells were examined with TdT modided biotin dUTP Nick end labeling(TUNEL)at different times. Results: ①In normal cochlear hair cells, no apoptic cell were generally noted. In 3 groups the morphological changes of the cell apoptosis were noted at 1,3 and 6 days after injection with amikacin; ②ABR threshold had no obvious changes 1,3 days after using amikacin while an elevation of ABR threshold was noted after using amikacin for 6 successive days in guinea pigs. Conclusion: ① Amikacin can induce the apoptosis of the cochlear hair cells in guinea pigs. Cell apoptosis was a mode of hair cell loss after injection with amikacin in guinea pigs. ② The ototoxin mechanism of the aminoglycoside antibiotic may be related to apoptosis of the cochlear hair cell induced by itself.
