摘要
目的观察白藜芦醇对肝癌细胞Bel-7404增殖、凋亡、侵袭影响,并研究其内在分子机制。方法采用MTT法检测白藜芦醇对Bel-7404细胞增殖的影响;流式细胞术检测白藜芦醇对Bel-7404细胞凋亡的影响;Transwell法检测白藜芦醇对Bel-7404细胞侵袭的影响。蛋白免疫印迹实验检测Pro-caspase 3,PARP,MMP-9及VEGF的表达。结果白藜芦醇可以抑制Bel-7404细胞增殖,呈时间和浓度依赖性。白藜芦醇可以诱导Bel-7404细胞凋亡,呈浓度依赖性,与下调Procaspase3和PARP蛋白表达有关。白藜芦醇可以抑制Bel-7404细胞侵袭性,呈浓度依赖性,与下调MMP-9、VEGF蛋白表达有关。结论白藜芦醇可以抑制肝癌Bel-7404细胞增殖、通过剪切Procaspase3,PARP蛋白而诱导凋亡,通过下调MMP-9和VEGF蛋白表达水平而抑制Bel-7404细胞侵袭性。
OBJECTIVE To observe the effect of Resveratrol on proliferation,apeptosis,invasion in liver cancer Bel-7404 cells and investigate the intrinsic molecular mechanism. METHODS MTT assay was used to evaluate the anti-proliferation effect of Resveration;Flow cytometry was carried on to detect the apoptosis induced by resveration; Transwell assay was used to detect the impact of resveration on invasion of ceils; Western bolt was carried on to detect the Pro-caspase 3,PARP, MMP9 and VEGF protein expression. RESULTS Resveration could inhibit proliferation of Bel-7404 cells in dose and time dependent manner; Resveration induced apeptosis of Bel-7404 ceils in dose dependent manner with down-regulated Pro-caspase 3 and PARP protein expression; Resveration inhibited invasion of Bel-7404 cells in dose dependent manner with decreased MMP-9 and VEGF protein expression. CONCLUSION Resveration could inhibite proliferation of Bel-7404 cells, induce apoptois through activate and shear procaspase3 and PARP-l,inhibit invasion through down-regulation of MMP-9 and VEGF.
