摘要
目的探讨高原习服过程中大鼠血浆瘦素(leptin)水平及弓状核内c-fos阳性神经元数目的变化。方法将48只健康成年SPF级雄性SD大鼠随机分为对照[西宁(海拔2 262 km)]组和第1、3、7、15、30天组[花石峡(海拔4 300 km)],每组8只。于饲养的既定时间检测血浆瘦素水平和弓状核中c-fos阳性神经元数目。结果与对照组比较,从急进高原第1天开始,各组大鼠血浆瘦素水平和下丘脑弓状核c-fos阳性神经元数目均升高,差异有统计学意义(P<0.01)。随着急进高原时间的延长,大鼠血浆瘦素水平和下丘脑弓状核c-fos阳性神经元数目均呈先上升后下降的趋势,且分别在第7、3天达到峰值。结论高原习服过程中大鼠血浆瘦素水平及弓状核内c-fos阳性神经元数目呈现先升高后降低的变化规律,推测血浆瘦素可能参与了高原习服不良的发生。
Objective To investigate the changes of plasma leptin and the c-fos positive neurons in arcuate nucleus in rats during the altitude acclimatization process. Methods A total of 48 healthy adult male SD rats were randomly divided into six groups:the control group[Xining(elevation 2 262 km)],the l st,3rd,7th, 15th and 30th day group [Huashixia (elevation 4 300 km)],eight each group. The plasma leptin levels and the c-fos positive neurons of hypothalamic arcuate nucleus of each group were detected at the given time. Results Compared with the control group,the plasma leptin levels increased significantly during the day of radical plateau (P〈0.01). Compared with the 1st day group,the plasma leptin levels increased significantly from the 3rd day to the 7th day (P〈0.01). Compared with the 3rd,7th day group,the plasma leptin levels decreased significantly in the 15th day group (P〈0.01), but still higher than those in the 1st day group (/:'〉0.05). The plasma leptin levels were further decreased in the 30th day group,and restored to the level of the 1st day group. The c-fos positive neurons in the arcuate nucleus increased during the radical plateau observed under the microscope. It was the highest in the 3rd day group and reduced gradually in the 7th day group,further reduced in the 15th,30th day group. Conclusion The changes of plasma leptin and the c-los positive neurons in arcuate nucleus in rats during the altitude acclimatization process show a low of increase first then decrease, based on this, plasma leptin may be involved in the occurrence of adverse altitude acclimatization.
