摘要
目的研制肺炎克雷伯菌(KP)全基因组DNA芯片并对芯片质量进行评价。方法根据KP的NTUH-K2044全基因组序列,挑选出5075条基因,PCR扩增各基因并纯化产物,点样制备芯片。设计了3个质控杂交组合,采用双色荧光杂交策略,对芯片质量进行评价。结果与结论芯片杂交结果与理论预期结果一致。本研究成功研制了KP全基因组DNA芯片,并建立了基于全基因组DNA芯片的KP比较基因组学技术平台。
Objective To develop and evaluate a whole-genome DNA mieroarray of Klebsiella pneumoniae. Methods Based on genomic sequences of K. pneumoniae NTUH-K2044, a total of 5075 genes were chosen, amplified by PCR and printed onto glass slides. Three sets of two-fluorescence comparative hybridizations were performed to evaluate the quality of the microarray. Results and Conclusion Microarray hybridization results were consistent with the anticipated results. The whole-genome DNA microarray of K. pneumoniae is successfully developed, and a method of microarray-based comparative genomic analysis of K. pneumoniae is established accordingly.
