摘要
10^(-10)和10^(-9)mol/L内皮素灌流离体大鼠心脏,抑制心肌摄取^(14)C-乳酸分别为38％和48％。在大鼠心肌肌膜蛋白重组脂质体模型上,10^(-10)和10^(-9)mol/L内皮素抑制其乳酸的跨膜载体转运分别为23％和33％。实验结果证实,内皮素能直接抑制心肌对乳酸摄取的跨膜载体转运,从而影响心肌的正常代谢方式。
Endothelin (ET) is a vasoactive polypeptide produced from the vascular endothelium with potentvasoconstrictive action. Under certain pathologic conditions it participates in the myocardial damageduring ischemia or anoxia, but its mechanism of action is not elucidated. This work is performed onperfused isolated rat heart. It is shown that the myocardial ^(14)C-labelled lactate uptake is 8.12±0.9μmol/g ww/10 min. When hearts are perfused with 10^(10) or 10^(-9) mol/L ET, the myocardial ^(14)C-lactateuptake is significantly inhibited, being 5.06±0.66 and 4.38±0.53 μmol/g ww/10 min (P<0.05 andP<0.01)respectively. On a model of liposome reconstituted with rat myocardial sarcolemmal protein,10^(-10) or 10^(-9) mol/L ET obviously inhibited transmembrane transfer of lactate through carrier transportsystem by 23% and 33% respectively. Results showed that ET directly inhibited the myocardial trans-membrane carrier transport system for lactate uptake, therefore interferes normal myocardial motabo-lism.
