摘要
为获得较好的制备杉木单细胞核悬浮液的试验方法,采用正交试验设计筛选其试验组合。杉木单细胞通过化学解离后,经过滤、离心再过滤得到细胞碎片少、粘连体少且细胞核完整的杉木单细胞核悬浮液;最优解离条件为:以WPB为解离液、解离时间4 h、离心转速800 r/min、离心时间10min;50μm过滤棉可显著降低粘连体及碎片的数量,有利于得到纯度高的单细胞悬浮液。
The orthogonal experiment design was adopted to select satisfactory method for intact Cunninghamia lanceolata cell nuclei suspension preparation. The intact cell nuclei suspension of Cunninghamia lanceolata with less cell fragments and joints could be obtained through chemical dissociation, filtration, centrifugation, and re-ill- tration. The experiments showed that the optimal dissociation conditions were taking 4 hr of dissociation, and 800 r/rain centrifugation for 8 min. Filtration with 50 μm reduce the cell fragments and joints and promote the purity of the intact cell nuclei WPB as dissociation solution, filter cotton could significantly suspension.
