摘要
目的用数字化基因表达谱分析益气活血方作用前后大鼠肺组织基因表达的变化,了解益气活血方作用的分子机制。方法 Wistar大鼠20只,随机分为中药组与对照组各10只。中药组大鼠每日灌胃益气活血方1 mL,对照组大鼠每日灌胃蒸馏水1 mL,两组均于6、12周末随机处死5只大鼠,取其右肺,并提取肺组织总mRNA,经过处理后进行数字化基因表达谱分析,对比两组基因差异表达情况。结果中药组、对照组在6周末的差异表达基因有1338个,上调基因为705个,下调基因为633个;上调基因主要与细胞分裂增殖相关,下调基因主要与脂代谢、炎症因子相关。12周末差异基因有2001个,上调基因为775个,下调基因为1226个。上调基因主要与免疫、DNA损伤修复相关,下调基因主要与脂代谢、炎症因子、纤维化功能相关。结论益气活血方可以调控脂代谢、炎症因子、免疫功能、DNA损伤修复、纤维化相关基因的表达,具有一定抗辐射功能,对机体有性保护作用。
Objective： To analysis differentially expressed genes of rat lung tissue before or after being given Yiqihuoxue Decoction by using the digital gene expression profiles,in orde to explore the mechanism of this recipe.Methods： 20 Wistar rats were randomly divided into the Chinese medicine group,control group.1 mL Yiqihuoxue Decoction were garaged to the Chinese medicine group,meanwhile 1 mL distilled water were gavaged to the control group.5 rats were random selected and killed at 6 th,12 th weekend after the first time gavaged,then the right lungs were taken,and total RNA of lung tissue was extacted.Digital gene expression profiling was analysised after treatment,and different gene expression betwwen two groups were compared.Result： There 1338 differentially expressed genes betwwen chinese medicine group and control group,of them up-regulated genes was 705,down-regulated genes was 633.The up-regulated genes were related to Cell division multiplication,and the down-regulated genes were related to Lipid metabolism and inflammation factor.There were 2001 differentially expressed genes between chinese medicine group and control group,of which up-regulated genes was 775,down-regulated genes was 1226.The up-regulated genes were related to Immunity and DNA damage repair,and the down-regulated genes were related to lipid metabolism,inflammation factors and fibrosis.Conclusion： Yiqihuoxue Decoction can regulate and control the exprssion of Lipid metabolism,inflammation factors,immune function,DNA damage repair and fibrosis related genes,and can resist radiation,and protect the organism.
