摘要
目的探讨脑源性神经营养因子(BDNF)基因多态性(Val66Met)与抑郁症及认知功能的关系。方法对符合纳入及排除标准的抑郁症患者组及健康对照组进行临床资料收集与血样采集。采用PCR-RFLP方法对BDNF基因多态性(Val66Met)进行分型,数据采用SPSS11.5软件进行统计学分析。结果中国陕西省汉族抑郁症患者BDN-FVal66Met基因多态性存在3种单倍型分布,分别为Val/Val、Val/Met和Met/Met,但与抑郁症不存在关联性;BDNF基因多态性(Val66Met)与患者WCST的分类数、语言流畅性的流畅总数和重复个数有关。结论 BDNF基因多态性(Val66Met)在陕西汉族抑郁症患者中的分布无差异,但该基因多态性可能与抑郁症患者的认知功能有关。
Objective To study the relationship of BDNF gene polymorphism(Val66Met) with major depression and cognitive functions.Methods Totally 185 patients and 199 normal controls were recruited in this study.Their clinical data and blood samples were collected.Of the patients 87 were measured for cognitive functions by verbal fluency test,Trail Making Test,WCST and WAIS-RC.The gene polymorphisms of BDNF were determined with PCR-RFLP technique.SPSS11.5 was used for statistical analysis.Results Three genotypes of BDNFVal66Met were detected in the depressed patients of Han nationalities in Shaanxi Province: Val/Val,Val/Met and Met/Met.No significant association was found between BDNF genotype and depression.However,BDNF genotype was associated with WCSTA,VFTAN and VFTR in the patients.Conclusion BDNF gene polymorphism does not play a major role in the pathogenesis of depressed patients.Val66Met may be associated with cognitive function of depressed patients.
