摘要
通过观察红豆杉提取物对急性心梗动物模型心肌细胞中Bcl-2、Bax凋亡因子的影响,探讨其对梗死心肌的保护作用及机制。采用结扎左冠状动脉分支法复制心肌梗死动物模型,运用免疫组化法定量分析心肌组织中Bcl-2和Bax表达的变化。结果表明:与模型组相比,红豆杉提取物给药组可增加心肌组织中Bcl-2的表达,抑制Bax的表达。红豆杉提取物可通过上调Bcl-2和下调Bax的表达,使Bcl-2/Bax值上升,抑制心肌细胞凋亡,从而对梗死心肌的保护起作用。
We studied the effect of the extract of Taxus cuspidata on Bcl-2 and Bax in myocardial cells of acute myocardial infarction animal model and on the protection of infarction myocardium and its mechanism.We adopted the myocardial infarction animal model of ligaturing the left branch of coronary attery,and quantitatively analyzed the change of the expression of Bcl-2 and Bax in myocardium with the means of immunohistochemistry.The results showed that the extract of Taxus cuspidata could obviously increase the expression of Bcl-2 in myocardium,and reduce the expression of Bax in myocardium.The extract of Taxus cuspidata could upregulate Bcl-2 gene expression and downregulate Bax gene expression and increase the ratio of Bcl-2 and Bax to restrain myocardial apoptosis and promote the infarction myocardium.
