摘要
目的观察灯盏花素对人肝癌细胞HepG2及人正常肝细胞LO-2体外增殖的影响。方法以不同浓度(0、3.125、6.25、12.5、25、50 mg.L-1)的灯盏花素分别处理HepG2和LO-2细胞,24 h后采用MTT法检测各组细胞生长情况;Hoechst 33258荧光法和流式细胞术观察HepG2和LO-2的凋亡情况。结果灯盏花素在3.1~50.0 mg.L-1范围内呈浓度依赖性地抑制HepG2细胞的增殖,24 h的IC50为8.4 mg.L-1,并能诱导HepG2细胞凋亡;但对LO-2的增殖影响较小,且几乎无诱导凋亡作用。结论灯盏花素体外可选择性地抑制肝癌细胞的增殖,可能与其诱导肝癌细胞凋亡有关。
Objective To investigate the effects of breviscapine on proliferation of human hepatocellular carcinoma cell line HepG2 and human normal hepatic cell line LO-2 in vitro.Methods HepG2 and LO-2 were incubated with various concentration(0,3.125,6.25,12.5,25,50 mg·L-1) of breviscapine for 24 hours respectively,and the cell proliferation was analyzed by MTT.Hoechst33258 fluorescence method and flow cytometry technique were adopted to analyze the apoptosis on HepG2 and LO-2.Results breviscapine displayed a cytotoxic response on HepG2 at(3.1~50.0) mg·L-1,dose-dependently,the IC50 value of HepG2 was 8.4 mg·L-1 and could induce the apoptonzsis of HepG2 cell line,but no LO-2.Conclusion The results suggest that breviscapine has antiproliferative effect on HepG2 in vitro,which may be related to inducing cell apoptosis.
