ALB、TLR7、PSPH和WSB1基因在雏鸭肝炎病毒易感与抗性群体间的差异表达分析 被引量:1

Comparison of ALB,TLR7,PSPH and WSB1 genes mRNA expression between susceptible and resistant populations to duck hepatitis virus

摘要 采用实时荧光定量RT-PCR技术检测雏鸭肝炎病毒侵染后对照组、易感组与抗性组ALB、TLR7、PSPH和WSB1mRNA分别在肝、脾、肺、肾、大脑、小脑、腿肌和胸腺等组织中的相对表达量并分析其意义。结果表明,除腿肌外,ALB和TLR7基因在易感组中的表达量极显著低于对照组和抗性组(P<0.01),抗性组的表达量显著或极显著低于对照组的表达量(P<0.01或P<0.05);PSPH和WSB1基因在易感组中的表达量极显著高于对照组(P<0.01),而抗性组与对照组差异不显著(P>0.05)。研究结果揭示了鸭ALB和TLR7基因为雏鸭肝炎病毒的抗性基因,其表达量的变化可以作为区分易感鸭和抗病鸭的标志。 This research detected the relative expression level of ALB, TLR7, PSPH and WSB1 genes in susceptible and resistant group infected by hepatitis virus and control group of duck- lings. The expression level of ALB, TLR7, PSPH and WSB1 genes in livers, pancreases, lungs, kid- neys, cerebra,cerebella, leg muscles and thymuses of different groups were detected by real-time quantitative RT-PCR. The results showed that except in leg muscles,the expression level of ALB and TLR7 genes in susceptible group is highly significantly less than that in resistant group and control group(P^0. 01) ,and the expression level in resistant group is highly significantly or sig- nificantly less than that in control group(P^0.01 or P^0.05). The expression of PSPH and WSB1 genes in susceptible group is highly significantly greater than that in control group (P~ 0.01),but the difference between resistant group and control group is not significant(P^0.05). This results is the same as that of suppression subtractive hybridization, and makes a further effort in revealing that the downregulators ALB and TLR7 are the resistant genes in ducklings hepatitis. And the change of the expression level of those two genes can be used as an indication to differ susceptible ducks and resistant ducks.
