摘要
目的建立松龄血脉康胶囊中葛根及马尾松鲜松叶的UPLC指纹图谱分析方法,为其质量评价累积数据。方法采用RP-HPLC法,以Agilent Eclipse Plus C18 RRHD色谱柱(150 mm×2.1 mm,1.8μm);流动相为0.1%甲酸(A)-乙腈(B),梯度洗脱时间60 min;检测波长250 nm;体积流量0.2 mL.min-1;柱温30℃;进样量1μL,进行指纹图谱研究。同时通过液相色谱-电喷雾离子阱质谱分析方法鉴别部分成分。结果建立了松龄血脉康胶囊的指纹图谱,以葛根素为参照峰,确定了28个共有峰,测定了32批样品,样品指纹图谱相似度均大于99%,共鉴别出25个相关成分。结论所建方法操作简便,分离效果好、稳定性高、重复性好,UPLC法使缩短了检测时间,可应用于松龄血脉康胶囊的质量控制及真伪品鉴别。
OBJECTIVE To establish the HPLC fingerprint of Songling Xuemaikang capsules and accumulate data for evaluation of its quality.METHODS The HPLC fingerprints were determined with RP-HPLC on Agilent Eclipse Plus C18 RRHD column(150 mm×2.1 mm,1.8 μm)with the gradient solvent system consisting of 0.1% formic acid-water(A) and acetonitrile(B).The flow rate was 0.2 mL·min-1,the detection wavelength was set at 250 nm,and the column temperature was maintained at 30℃.The data was analyzed using 'similarity of Chinese traditional medicine fingerprints' software.UPLC tandem ESI Linear Ion trap Mass Spectrometer was used to identify the components of Songlingxuemaikang capsules.RESULTS Taken Puerarin as reference peak,thirty-two batches were analyzed,which 25 major components were identified with high similarity of fingerprints.CONCLUSION This method is easy to handle and has good separation,reproducibility and stability.Due to the application of UPLC technology,the assay time is shorten to only 60 min.It can be applied for quality control and product identification of Songling Xuemaikang capsules.
