牛黄参含药血清诱导大鼠肝干细胞分化及调控机制的研究

摘要 目的:研究牛黄参含药血清对大鼠肝干细胞系WB-F344增殖、分化及凋亡的影响,探讨其作用机制。方法:MTT法检测不同浓度含药血清对WB-F344增殖的影响;取增殖的最佳浓度,Westernblot检测含药血清对WB-F344细胞因子HGFR、EGF、EGFR、TGFβ1R、TGFβ2R表达的影响;流式细胞仪检测含药血清对WB-F344细胞凋亡的影响。结果:MTT结果显示10%、20%浓度的含药血清均能促进WB-F344细胞的增殖,且以10%为增殖的最佳浓度(P<0.01);Westernblot结果显示经10%的含药血清干预后,WB-F344细胞HGFR、EGF、EGFR、TGF-β1R、TGF-β2R的表达量上调(P<0.01);流式细胞仪检测结果则显示含药血清干预后WB-F344细胞凋亡率显著下降(P<0.01)。结论:牛黄参含药血清可促进大鼠肝干细胞的增殖、分化,并抑制其凋亡,这可能是其抗肝损伤、促进肝再生的作用机理之一。 Objective:To observe the effects of tonifying qi and detoxication prescription(niuhuangshen) contained serum on rats’ liver stem cell line WB-F344 proliferation, differentiation and apoptosis, explore the mechanism. Methods:The effects of different concentrations of niuhuangshen contained serum on proliferation of WB-F344 were determined by MTT colorimetry;selecting the optimal concentration of niuhuangshen contained serum on proliferation, the effects of niuhuangshen contained serum on the expressions of HGFR, EGF, EGFR, TGFβ1R and TGFβ2R in WB-F344 cells were detected by Western blotting, WB-F344 cells apoptosis was determined by flow cytometer . Results:MTT results showed 10% and 20% concentration of niuhuangshen contained serum could promote proliferation of WB-F344, the optimal concentration for proliferation was 10% (P<0.01); Western blotting results showed that niuhuangshen contained serum could up-regulate the expression HGFR, EGF, EGFR, TGFβ1R and TGFβ2R in WB-F344 cells(P<0.01); and apoptosis rate of WB-F344 cell significantly decreased after intervened by drug-containing serum (P<0.01). Conclusion:The prescription of tonifying qi and detoxication(niuhuangshen) contained serum can promote rats’ liver stem cell proliferation, differentiation, and inhibiting its apoptosis to stimulate regeneration of liver cell by acting on cytokine system.
