摘要
本试验以甜高粱秸秆汁为主要的发酵原料,应用二次正交旋转组合试验设计方法对热带假丝酵母SG-1产单细胞蛋白的发酵培养基进行优化。结果显示,热带假丝酵母SG-1产单细胞蛋白的较佳发酵培养基组成为:甜高粱秸秆汁220 mL/L、工业葡萄糖37.5 g/L、工业蛋白胨37.5 g/L、MgSO4.7H2O 3.75 g/L、KH2PO43.75 g/L。该优化条件下单细胞蛋白产量达15.38 g/L,比初始发酵培养基(甜高粱秸秆汁120 mL/L、工业葡萄糖20 g/L、工业蛋白胨20 g/L、MgSO4.7H2O 2.0 g/L、KH2PO42.0 g/L)单细胞蛋白产量(10.77 g/L)提高了42.80%。
In this paper,using the sweet sorghum stalk juice as the major fermentation substrate,the fermentation medium for SCP-producing strain Candida tropicalis SG-1 was optimized with the method of rotational quadratic orthogo- nal experiment design.As a resuh,the optimal fermentation medium was as follows: sweet sorghum stalk juice 220 mL/L, glucose 37.5 g/L, peptone 37.5 g/L, MgSO,. 7H2O 3.75 g/L, KH2PO4 3.75 g/L. Under the optimal fermentation medium,the final SCP yield reached 15.38 g/L,which was 42.80% higher than that obtained with the original medium recipe (sweet sorghum stalk juice 120 mL/L, glucose 20 g/L, peptone 20 g/L, MgSO4· 7H2O 2.0 g/L, KH2PO4 2.0 g/L).
