癫痫鼠痫性皮层及正常鼠脑皮层组织基因的差异表达研究 被引量:1

摘要 目的:探究癫痫鼠痫性皮层及正常鼠脑皮层组织基因的差异。方法:本实验将40只大鼠随机分为癫痫组与对照组2组,其中电刺激一侧杏仁核点燃20例作为模型组,放置刺激电极但未予电刺激点燃(假手术组)20例作对照组。采用DDRT-PCR技术研究癫痫鼠痫性皮层和正常鼠脑皮层组织基因的差异。结果:研究癫痫鼠痫性皮层和正常鼠脑皮层组织基因的差异,从中发现了7个差异基因.Timm23,NGF-IA,Fbx38_predicted,ATPase synthase,subunit 6,APP,PAF-AH beta,NG5 protein。其中3个基因高表达,4个基因低表达。结论:采用DDRT-PCR技术有助于快速发现了癫痫鼠痫性皮层和正常鼠脑皮层组织差异的7个差异基因,这些基因有可能与癫痫产生有关。 Objective:Study on the difference of the genes between the epileptic rats and the normal ratsapos;cerebral cortex tissues.Methods:In this experiment,40 rats were randomly divided into 2 groups:the epilepsy group and the control group.The 20 cases of electrical stimμlation of the unilateral of the amygdala were considered as the model group.Another 20 cases placed a stimμlation electrode but without electrical kindling stimμlations(sham operation group) were served as the control group.By using DDRT-PCR technology we studied the deferent expression of the genes between the epilepsy ratsapos;epileptic cortex and normal ratsapos;cerebral cortex tissue.Results:The study found 7 different genes.They are Timm23,NGF-IA,Fbx38 predicted,ATPase synthase,subunit 6,APP,PAF-AH beta,NG5 protein.Three genes show high expression.Four genes show low expression.Conclusion:The DDRT-PCR technique contributes to the rapid discovery of the deferent expression of seven genes epilepsy ratsapos;epileptic cortex and normal ratapos;s cerebral cortex.These genes has the relation with the epilepsy rats.
