摘要
目的:筛选五味子-细辛药对抗哮喘的活性部位。方法:采用氯化乙酰胆碱磷酸组胺引喘法、小鼠氨水引咳法和气管酚红排泌法,观察该药对5种提取物(五味子挥发油、五味子水提液、五味子总酸、细辛挥发油及细辛水提液)对豚鼠的平喘、小鼠的镇咳和祛痰作用。结果:豚鼠分别ig五味子挥发油、细辛挥发油,豚鼠哮喘潜伏期显著延长,其它3种提取物均无显著作用;小鼠分别ig五味子挥发油、五味子水提液、五味子总酸、细辛挥发油,小鼠咳嗽潜伏期显著或极显著地延长,咳嗽次数极显著地减少,细辛水提液无显著作用;小鼠分别ig五味子挥发油、五味子水提液、五味子总酸、细辛挥发油,小鼠气管酚红排泌量均极显著地增多,细辛水提液无显著作用。结论:该药对的平喘活性部位可能为五味子挥发油和细辛挥发油,镇咳及祛痰活性部位可能为五味子挥发油、五味子水提液、五味子总酸、细辛挥发油。
Objective：To screen the anti-asthma bioactive extracts for the drug partnership containing fructus schisandrae and asarum.Methods：The asthma suppression effects of five extracts（the volatile oil,water extract and crude free organic acid from fructus schisandrae the volatile oil,water extract from asarum） were studied by asthma latent period in guinea pigs induced by the acetylcholine chloride and histamine phosphate.The antitussive activities of this five extracts were observed through the pre-cough time and cough times in mice by the ammonium hydroxide.Their expectorant activities were identified by determining the excreting amount of the phenol red in mice.Results：The volatile oils both from fructus schisandrae and asarum were significantly prolonged the pre-convulsive time（PCT） of guinea pigs induced by the mixture spray of the histamine and acetylcholine chloride（P0.05）,while the other three extracts took no significant effect.The volatile oil,water extract and crude free organic acid from fructus schisandrae as well as the volatile oil from asarum were respectively able to prolong the cough latent period（P0.05 or P0.01 ）and decrease the cough frequency （P0.01 ）in mice caused by the ammonia liquor,however the water extract from asarum had no significant antitussive activity.The tracheal phenol red excretion in mice was dramatically enhanced by the volatile oil,water extract and crude free organic acid from fructus schisandrae as well as the volatile oil from asarum respectively（P0.01）,but the water extract from asarum took no significant effect.Conclusions：The asthma suppression extracts of the drug partnership containing fructus schisandrae and asarum may be the volatile oils both from fructus schisandrae and asarum.The antitussive and expectorant extracts of this drug partnership were possibly the volatile oil,water extract and crude free organic acid from fructus schisandrae as well as the volatile oil from asarum.
