摘要
为应用耐寒常春藤的有利性状,以嫩茎为材料,采用生根培养的方法,对其进行了生根培养、继代扩增培养以及移栽和移植的研究。结果表明:把无菌嫩茎的下部切口在浓度为10mg.L-1的IAA溶液中处理3min后,接种到1/2MS+蔗糖20g.L-1+NAA0.2mg.L-1~0.4mg.L-1的培养基上是耐寒常春藤无菌嫩茎生根培养的理想培养方法;把试管苗剪成具有侧芽或顶芽的茎段后,在1/2MS+蔗糖20g.L-1+NAA0.3mg.L-1的培养基上进行扩增培养是耐寒常春藤试管苗扩增培养的理想方法;试管苗移栽成活率在95%以上;定植2年后陆地越冬成活率在91%以上;试管苗保持了耐寒变异性状。
In order to preserve the wild resources of the cold--resistant Hedera nepalensis K,this article uses tender stems of Hedera nepalensis K as explants to do rooting cluture, and proliferates cultivation of roots and transplanting. The results indicate that the ideal medium for rooting of tender stems is 1/2 MS+ sucrose (20goL^-1)+NAA(0.2mg ·L^-1-0.4mg ·L^-1) with the method that the lower incision of sterile stems im merse at the 10mg ·L^-1 IAA solution in 3 minutes; after the seedlings are sheared into stems with lateral buds or terminal buds,the perfect medium for proliferating cultivation of them is 1/2MS+ sucrose30 g/L + NAA 0.3 mg/L; the transplanting survival rate is above 95 % ;after planting for two years the survival rate of the overwintering plants is above 91% and the test--tube plantlets maintain a cold--resistant characteristic.
