摘要
目的 建立和优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术 ,初步观察巴豆提取物诱导小鼠小肠组织中蛋白质的差异表达 ,为进一步筛选其中介导巴豆生物作用的蛋白质分子奠定基础。方法 应用巴豆提取物灌胃制备小鼠慢性胃肠运动增强模型 ,提取其小肠组织中总蛋白质 ,优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术 ,并用其分析模型动物小肠组织中蛋白质 ,双向电泳凝胶银染法显示其蛋白质的差异表达。结果 建立了小鼠慢性胃肠运动增强模型 ,优化并运用双向聚丙烯酰胺凝胶电泳技术展示出模型动物小肠组织中有差异蛋白质的表达。结论 巴豆提取物可以诱导小鼠小肠组织中蛋白质差异表达 。
Aim To establish and optimize the two dimensional(2 D)polyacrylamide gel electrophoresis(PAGE)for studying the different expression of small intestinal proteins induced by extract of croton(croton oil,CO) in BALB/c mice,and lay the foundation for screening and characterization of biological molecules mediating the effect of croton from the gastrointestinal tract.Methods To establish the model of chronic gastrointestinal motility hyperfunction in mice induced by intragastric administration of CO,extracting the proteins of small intestine from model mice.Optimizing the 2 D PAGE for analyzing the different expression of proteins.The 2 D PAGE gels were stained with silver staining.Results Animal model of chronic gastrointestinal motility hyperfunction was established in mice.Optimized 2 D PAGE has displayed the different expressional proteins in gastrointestinal tract of model mice.Conclusions The different expression of intestinal protein can be induced by extract of croton in BALB/c mice.These proteins may be medicate the effect of croton in gastrointestinal tract.
