摘要
为增强猪支原体肺炎活疫苗在肌肉注射免疫时的细胞免疫刺激能力,对7种猪支原体肺炎活疫苗可用佐剂的细胞免疫应答促进作用进行了体外及体内试验评价。7种佐剂中除皂角素外,其余6种对活疫苗活力皆无明显影响。用这6种佐剂体外刺激淋巴细胞,发现左旋咪唑、免疫刺激复合物基质、黄芪多糖和胞壁酰二肽可以在体外单独刺激或协同刀豆蛋白刺激小鼠脾淋巴细胞及猪外周血淋巴细胞增殖,其中免疫刺激复合物基质仅需ng/ml级别浓度即可起效,而壳聚糖和卡波姆不能在体外刺激细胞增殖。选用其中5种佐剂与活疫苗配合肌肉注射免疫小鼠,发现左旋咪唑、免疫刺激复合物基质、黄芪多糖、卡波姆可以显著增加机体针对猪肺炎支原体抗原的细胞免疫应答。结果表明,利用适当佐剂可以实现猪支原体肺炎活疫苗在肌肉注射条件下对机体细胞免疫应答的有效刺激。
To achieve an effective cellular immune stimulation by the attenuated live vaccine against Mycoplasma hyopneumoniae of swine when inoculated intramuscularly,different adjuvants were tested for their abilities to stimulate lymphocyte by in vitro and in vivo assays.Among all seven adjuvants,six of them,expect for saponin,showed no obvious toxicity to the live vaccine.In vitro stimulation assay revealed that levamisole,ISCOM-matrix,astragalus polysaccharide and muramyl dipeptide significantly stimulated the proliferation of mice spleen lymphocyte and swine peripheral blood mononuclear cell both by itself and with Con A.Chitosan and carbomer did not exhibit any effective in vitro stimulation.In the in vivo experiment,mice intramuscularly immunized with vaccine combined with levamisole,ISCOM-matrix,astragalus polysaccharide or carbomer showed significant increase of specific cellular immune response compared with the mice immunized with live vaccine only or PBS.In conclusion,the effective cellular immune response by intramuscular immunization of live vaccine against M.hyopneumoniae of swine can be achieved by using suitable adjuvant,which will help the subsequent development of the intramuscular inoculation of this live vaccine.
