摘要
目的:探讨沙土鼠脑缺血时中缝背核神经元5-HT的变化。方法:用免疫组织化学ABC法及图象分析对沙土鼠脑缺血不同时间以及缺血10分钟后再灌不同时间其中缝背核神经元5-HT的变化进行研究。结果:脑缺血10分钟、30分钟时,沙土鼠脑中缝背核5-HT阳性神经元的平均光密度值(OD值)与对照组比较无显著性差异;而缺血4小时时则明显降低,差异有显著性(P<0.05);脑缺血10分钟后,再灌流30分钟,5-HT下降不明显,而4小时时则明显降低(P<0.05),至24小时时达最低水平(P<0.05),36小时逐渐恢复正常。结论:当缺血至一定时间或缺血10分钟再灌流到一定时间时,中缝背核神经元内5-HT的含量减少。
Objective:To study the changes of 5-HT in nucleus raphe dorsalis in gerbils after cerebral ischemia and reperfusion. Methods: ABC methods of immunohistochemistry and image analysis technique were used to study the changes of 5-HT in nucleus raphe dorsalis after cerebral ischemia and reperfusion in different time. Results:The mean value of optic density of 5-HT im-munoreaction cell in nucleus raphe dorsalis was not sigmificantly different compared with control when cerebral ischemia lasted for 10 and 30 minutes. But when cerebral ischemia lasted for 4 hours, 5-HT contents of immunoreaction cell were decreased significantly. Thirty minutes reperfusion after 10 minutes cerebral ischemia,5-HT was not decreased significantly. However, after 4 hours reperfusion,5-HT was decreased significantly and reached the lowest after 24 hours reperfusion. After 36 hours reperfusion, 5-HT contents became normal gradually. Conclusion; When cerebral ischemia last for some time and reperfusion is long enough, content of 5-HT in nucleus raphe dorsalis is decreased.
