摘要
通过采用无菌发酵滤液梯度稀释、添加消泡剂对比试验和葡萄糖单因子试验,初步研究了海洋芽孢杆菌B-9987发酵过程中活菌量大幅波动的原因,为建立稳定的发酵工艺提供参考依据。结果表明,菌体胞外代谢产物和发酵过程中添加的消泡剂对菌体的生长具有抑制作用,培养基中葡萄糖含量也会引起菌体量波动,因此推测,三者共同作用导致发酵过程中活菌量大幅波动。通过综合调控培养基中葡萄糖含量和消泡剂的添加方式,解决了发酵过程中活菌量大幅波动的问题,并使发酵液中活菌量大幅提升,发酵结束时活菌含量在5.0×10^9cfu/mL以上。
The reasons of cell concentration drastic fluctuation during Bacillus marinus B-9987 fermentation was preliminarily studied,through the sterile filtrate dilution assay,adding defoamer contrast test and glucose single factor test. The results showed that extracelluiar metabolite and defoamer were harmful to the growth of Bacillus rnarinus 13-9987. The glucose in the medium could also cause fluctuations of the biomass concentration. Therefore,it is inferred that the com- bined action of the three factors resulted in cell concentration fluctuating substantially during the fermentation process. By adjusting the glucose content and defoamer-adding way during the fermentation process,which relieved the cell concentration fluctuating,and improved it at the same time, the cell concentration at the end of fermentation was as high as 5 × 10^9 cfu/mL.
