摘要 目的 扩增与人成骨肉瘤细胞特异结合的鼠源性单克隆抗体轻链基因 ,并对其基因序列进行分析 .方法 用RT- PCR方法从分泌鼠源抗人成骨肉瘤单克隆抗体的杂交瘤细胞 ( 7F6 B)扩增出抗体轻链基因 ,克隆到 p GEM- T载体 ,以全自动测序仪测定其轻链基因序列并对其结构进行分析 .结果  7F6 B轻链基因由 6 5 6 bp组成 ,编码 2 18个氨基酸 ,序列分析证实该基因可变区序列符合小鼠 Ig轻链基因的氨基酸序列特征 .结论 获得的鼠源性抗成骨肉瘤抗体轻链基因为κ 家族 。 AIM To amplify and analyze the light chain gene fragments of mouse anti-osteosarcoma monoclonal antibody. METHODS Light chain gene fragments of mouse antiosteosarcoma monoclonal antibody were amplified by RTPCR method using cell total RNA purified from antibody producing hybridoma 7F6B cells, and further cloned into pGEMT vector which was then sequenced. RESULTS 7F6B light chain gene fragments consisted of 656 bp and encoded 218 amino acid residues and the amino acid sequences of variable region were in agreement with the characteristics of the amino acids present in the mouse Ig variable region. CONCLUSION Light chain gene fragments of mouse antiosteosarcoma antibody belongs to κ light chain subgroup Ⅲ and successful cloning of this gene fragment might be used for further experiments of immunotherapeutic study of osteosarcoma.
