摘要
目的 观察补体调节蛋白 CD5 9和 CD46 在创伤性急性肺损伤 ( AL I)和急性呼吸窘迫综合征 ( ARDS)动物肺组织中的表达变化 ,并探讨其在 AL I和 ARDS发病机制中的意义 .方法 采用右胸撞击建立创伤性 AL I和 ARDS模型 .用免疫组化和原位杂交技术检查 AL I和 ARDS家猫肺组织中 CD5 9,CD46 的表达情况 .结果 伤后 12 h AL I动物肺组织中 CD5 9,CD46 表达上调 ,2 4h最为明显 ,48~ 72 h减弱 .ARDS动物肺组织中 CD5 9,CD46 表达明显下调 .结论 伤后早期 ,CD5 9,CD46 表达上调 ,加强了组织细胞对补体攻击的自我保护机制 ;ARDS时 ,CD5 9,CD46 表达下调可能是炎性损伤加重的原因之一 .
AIM To investigate the expression of CD 59 and CD 46 in the lungs of the cats with acute lung injury (ALI) or acute respiratory distress syndrome (ARDS) induced by blunt thoracic trauma. METHODS The animal model for ALI or ARDS was induced by impact on right chest wall of cats. The expression of CD 59 and CD 46 was determined by immunohistochemical staining and in situ hybridization. RESULTS The results showed the increased expression of CD 59 and CD 46 in the lungs of ALI cats during the period from 12 to 48 hours posttrauma and the decreased expression of CD 59 and CD 46 in the lungs of ARDS cats. CONCLUSION Inflammation in ALI may induce upregulation of CD 59 and CD 46 expression which protect the positive cells from attack of complement, and as inflammation develops into ARDS stage, downregulation of CD 59 and CD 46 expression may contribute to cells injury.
