外源性瘦素在急性肺栓塞合并肺损伤中的作用机制研究 被引量:6

摘要 目的通过观察外源性瘦素对兔急性肺栓塞(APE)后TNF-α、IL-6及IL-8水平的影响,探讨其在肺损伤时的保护机制。方法选取健康新西兰兔30只,随机分为三组,对照组、模型组、瘦素预处理组。用ELISA方法检测血清,术毕行病理分析。结果①瘦素预处理组,TNF-α、IL-6、IL-8水平比较生理盐水模型对照组明显下降,P值<0.05,差异具有统计学意义;生理盐水模型与对照组相比,数值明显高于对照组。②病理所见缺血渗出明显轻于模型对照组。结论外源性瘦素通过对急性肺血栓栓塞症后TNF-α、IL-6和IL-8的水平下调,从而对损伤的肺组织起到了保护作用。 Objective To investigate the effects of exogenous leptin on the levels of TNF-α,IL-6,IL-8 in rats with acute pul-monary embolism( APE), and explore its protective mechanism in lung injury. Methods Total 30 New Zealand rabbits were se-lected and randomly divided into 3 groups, the control group, model group and leptin group. The serum levels of TNF-α, IL-6 and IL-8 were tested by using ELISA,and the tissues were analyzed with pathological technology. Results ①The levels of TNF-α, IL-6 and IL-8 in the leptin group were higher as compared to the model group and control group( P 〈 0.05 ) ; And the levels in the model group were higher than those of the control group. ②The ischemia induced plasma leakage in the leptin group was light as compared to the control and model groups. Conclusion The exogenous leptin can protect the injured lung tissues by down-regu-lated the levels of TNF-α, IL-6 and IL-8 in rats wit APE.
