摘要
目的 :建立人子宫内膜癌裸鼠移植瘤株 ,为开展子宫内膜癌的实验性研究奠定基础。方法 :将 3例子宫内膜癌手术标本移植于裸鼠皮下 ,生长后行鼠间连续传代 ,对移植瘤进行相关的生物学检测。结果 :3例标本 1例获得生长并行鼠间传代 ,历时 2 4个月建成人子宫内膜癌裸鼠移植瘤株EnCa 95V ,瘤株生长稳定 ,已传 2 1代 ,移植成瘤率 10 0 % ,光镜、电镜、免疫组化、流式细胞术检测移植瘤与患者原始肿瘤特征基本一致 ;染色体分析显示人类肿瘤染色体核型 ,众数为 4 3~ 92条。结论 :EnCa 95V是一株新的子宫内膜癌裸鼠移植瘤株 。
Objective:In order to establish a xenografted tumour strain of human endometrial carcinoma in nude mice to provide a model in vivo for further investigation of this tumour.Metheods:Three human endometrial carcinomas derived from surgical specimens were transplanted into the nude mice.One graft was successively passaged after growing.A series of characteristics of the grafts were examined.Results:One of three tumors could be maintained by serial transplantation.The xenograft had been successively passaged 21 generations during a period of 24 months.A tumor strain EnCa95V was established,having a high rate of successful inoculation rate(100%) and growing steadly.The morphological and biolgical characteristics of its original tumor were preserved,The chromosomal analysis revealed an aneuploid human karyotype of 21~136 in number.The tumor markers and P 53 oncogenic product by immunocytochemical techniques showed high expression.Conclusions:The results showed that EnCa95V could be considered as a new xenograted tumor strain and may provide a model in vivo for further investigation of human endometrial carcinoma.
