摘要
目的 :探讨高温致神经管畸形 (NTD)的机制。方法 :利用高温致神经管畸形动物模型 ,在神经管发育的不同阶段 ,用免疫组织化学法检测神经生长因子 (nervegrowthfactor,NGF)和NGF受体 (trkA)在神经上皮、基底膜、脊索和神经管周围间充质中的分布状况 ,并观察高温对NGF及其受体的影响。结果 :NGF及其受体广泛分布于神经管及其周围组织 ,并随胎龄增加而呈规律性变化。高温处理后的胚胎神经上皮及其周围间充质中NGF及其受体的免疫组化着色强度明显减弱 ,着色细胞数明显减少。结论 :NGF及其受体的减少 。
Objective:To study up on the development mechanism of neural tube defects(NTDs) induced by hyperthermia.Methods:On the hamster NTD model,the distribution of NGF and its receptors(trkA) in the embryonic neural epithelium,its basement membrane,notochord and the mesenchyme around the neural tube was observed at different stages during neural tube development with the method of immunohistochemistry,and the effect of hyperthermia on the distribution and quantity of NGF and trkA was also analysed.Results:NGF and its receptors abundantly distributed in the neural tube and the mesenchyme in control groups.With the development of embryos,the expression of NGF and trkA changed ruly.The stained intensity for NGF,trkA in each treated group is weaker than that in control groups.The population of immunochemically stained cells markedly decreased in treated groups.Conclusion:The results may suggest that the reduction of NGF and trkA is an important aspect in NTD development caused by hyperthermia.
