摘要
目的 :探讨大鼠生后下丘脑不同发育阶段细胞凋亡的规律及禁水对细胞凋亡的影响。方法 :取生后发育不同阶段的大鼠下丘脑 ,①用Bouin液固定 ,用TUNEL染色 ;②用改良的Bouin液固定 ,核固红 结晶紫染色。结果 :①生后 1d凋亡细胞最多 ,主要位于下丘脑背侧室旁核附近。生后 5d ,凋亡细胞明显减少。以后凋亡细胞越来越少。 60d极少见到凋亡细胞。两种染色方法结果相似 ;②成年大鼠禁水后 ,未见凋亡细胞增多。结论 :①细胞凋亡受基因和细胞外环境因素的共同调节 ;
Objective:To study the rules of cell apoptosis in rat hypothalamus during the development of postnatal life and observe the effect of dehydration on cell apoptosis in adult rat hypothalamus.Method:Wistar rat hypothalamus was resected at different stages during the postnatal life and ① fixed by Bouin′s fixative and stained with terminal deoxyUTP nick end labelling technique(TUNEL)and ② fixed with modifide Bouin′s solution and stained with nuclear fast redcrystal violet.Results:①The rules of apoptosis:a peak density of apoptotic cells appeared on the first day (1d) after birth chiefly in the dorsal area of the paraventricular nucleus.On 5d the apoptotic cells reduced greatly,then became fewer and fewer,and almost disappeared on 60d.Similar results were produced by staining in two ways.②In adult rats deprived of water at different times,the number of apoptotic cells in hypothalamus was similar to that in controlled groups.Conclusion:①The apoptotic cells were controlled by genes together with pericellular environmental factors.②Dehydration had no effect on apoptosis.
