摘要
目的探讨轻型、重型聚丙烯疝网片对大鼠术后生殖细胞凋亡的影响。方法选取雄性成年SD大鼠32只,随机分成四组:空白对照组、手术对照组、轻型聚丙烯网片组(轻型组),重型聚丙烯网片组(重型组)。对后三组制作腹股沟疝模型,测定各组大鼠血液中MDA(丙二醛),睾丸组织中SOD(超氧化物歧化酶)、GSH(谷胱甘肽)含量,TUNEL法检测细胞凋亡。结果 4周时测定各组血清中MDA含量、睾丸组织SOD含量、GSH含量,收取睾丸组织标本制作石蜡切片,测定细胞凋亡数量(AI)。各组血清中MDA含量、睾丸组织SOD含量、GSH含量、AI手术对照组与空白对照组差异无统计学意义(t=0.652、0.246、0.177、0.944,P均<0.05),轻型组、重型组与手术对照组比较差异均有统计学意义(F=52.605、98.058、85.493、413.287,P均<0.05)。MDA含量与细胞凋亡指数,重型组高于轻型组,差异有统计学意义(t=4.944、8.122,P均<0.05);SOD含量与GSH含量,重型组低于轻型组,差异有统计学意义(t=5.398、4.643,P均<0.05)。结论无张力疝修补术后可引起大鼠血清MDA增加,睾丸组织中SOD、GSH含量减少,无张力山修补可引起大鼠生殖细胞凋亡。
Objective To explore the impacts on germ cell apoptosis after operation by using the light and heavy polypropylene hernia meshes. Methods Thirty-two adult male SD rats were randomly classi- fied into four groups including the control (blank) group, control (surgery) group,the group of light-weight polypropylene mesh (light group), the group of heavy-weight polypropylene mesh (heavy group). The ingui- nal hernia model were used for the last three groups and the MDA (malondialdehyde), SOD in testieular tis- sue,GSH (glutathione) and cell apoptosis by TUNEL method were determined. Results The contents of MDA, SOD, GSH were examined, the samples for histological test were collected and the number of cell apop- tosis (AI) was tested. No significant differences in MDA, SOD, GSH and AI were observed between the con- trol (blank) group and the control (surgery) group(t =0.652,0.246,0. 177,0.944 ,all P 〈0.05) ,the light group and the heavy group had significant difference than the control (surgery) group( F =52.605,98. 058, 85. 493,413. 287, all P 〈 0.05 ). The heavy group had significant more MDA and AI than the light group( t = 4.944,8. 122, all P 〈 0.05 ). While the SOD and GSH were observed to be lower in the heavy group com- pared to the light group(t = 5. 398,4.643, all P 〈 0.05). Conclusions The tension-free hernia repair can lead to an increase in serum MDA and decreases in SOD and GSH. The repair is also the reason for cell apoptosis in rats.
