摘要
目的探讨迷走神经在胸腺细胞成熟发育和输出过程中的作用。方法实验采取颈部右侧迷走神经切断术,应用免疫组织化学和流式细胞术检测大鼠胸腺CD4、CD8阳性细胞表达及分布变化情况。结果 CD4、CD8阳性细胞分布于大鼠胸腺特定区域,单侧迷走神经切断后阳性细胞分布面积减小,未成熟胸腺细胞百分比降低,成熟胸腺细胞百分比升高。结论迷走神经切断术后,胸腺中未成熟T细胞减少,成熟T细胞数量相对增多。迷走神经在T细胞发育及输出过程中起重要作用。
Objective To investigate the mechanism of vagus in regulating the migration of thymocytes in rat thymus. Methods In this study, immunohistochemistry and flow cytometry (FCM) was used. Monolateral vagal denervation was performed at the cervical level. The distribution of CD4/CD8 reactive ceils were investigated in rat thymus, as well as changes in the distribution of thymocytes subsets in rat thymus after monolateral vagotomy. Results CD4 and CD8 labeled cells were located at the corticomedul!ary junc- tions and medullas. After monolateral vagotomy, both the area and the percent of the CD4/CD8 positive stained cells were decreasing, while the percent of the mature thymocytes were increasing. Conclusion In rat thymus, monolateral vagotomy results in the decrease of immature thymocytes and the increase of mature thymocytes in number, which shows that the right vagus denervation produces a decrease of lymphoeytes release from the thymus into the venous circulation.
