摘要
目的研究狼疮肾炎(lupus nephritis,LN)患者跨膜蛋白活化物及钙调亲环素配体相互作用分子(TACI)的表达及其与疾病活动度的关系。方法分别采用逆转录实时荧光定量聚合酶链反应(FQ-RT-PCR)方法,检测42例LN患者,30例非LN患者,40例健康体检者外周血单个核细胞(PBMC)TACI mRNA表达水平;采用流式细胞术(FCM)检测外周血B淋巴细胞表面TACI表达百分率;采用酶联免疫吸附测定(ELISA)检测血清中抗双链DNA抗体(dsDNA)水平;采用速率散射比浊法检测血清中补体C3的水平。结果①LN组非LN组和和健康对照组TACI mRNA表达水平分别为:6.07±0.46,7.73±0.60和8.02±0.51。LN组TACI mRNA表达水平高于非LN组及健康对照组;非LN组TACI mRNA表达水平高于健康对照组(P<0.01)。②LN组、非LN组和健康对照组B淋巴细胞表面TACI表达百分率分别为:(7.95±1.58)%,(3.98±1.29)%和(2.87±0.56)%。LN组B淋巴细胞表面TACI表达百分率显著高于非LN组和健康对照组;非LN组高于健康对照组(P<0.01)。③LN患者PBMC中TACI mRNA的表达水平与抗dsDNA抗体呈显著正相关(r=0.312,P<0.05);与补体C3呈显著负相关(r=-0.315,P<0.05)。结论 LN患者TACI表达水平升高,可能参与了LN的发病过程;治疗后TACI表达水平降低,减轻炎症损害,可能成为评价疗效和预后的新指标。
Objective The objective of this study was to explore the association between the expression of transmembrane activator and calcium modulator and cyclophilin ligand interactor（TACI） receptor in patients with lupus nephritis（LN）. Methods Reverse transcription-real time quantitative polymerase chain reaction（FQ-RT-PCR） was used to examine TACI mRNA expression in peripheral blood monocytes of 42 cases of LN,30 cases in non-LN,and 40 healthy controls. TACI receptor percentage on B lymphocytes was measured by flow cytometry（FCM）. The anti- dsDNA antibody level in serum was assayed by ELISA. The serum complement C3 level was assayed by automatic immune analysis machine. Results （1）The TACI mRNA level in patients with LN,n0n-LN group and normal controls was 6.07±0.46,7.73±0.60 and 8.02±0.51. The TACI mRNA level in patients with LN was higher than that of non-LN group and normal controls, the TACI of non-LN group was higher than that of normal controls, there were significant difference among three groups（ P 〈0.01）. （2）The percentage of TACI on B lymphocytes in patients with LN, non-LN group and normal controls was （7.95 ±1.58）%, （3.98± 1.29）% and （2.87± 0.56）%. The percentage of TACI on B lymphocytes in patients with LN was higher than that of non-LN group and normal controls,the percentage of non-LN group was higher than that of normal controls,there were significant differences among three groups（ P 0.01）. QThe expression level of TACI mRNA in PBMC of patients with LN was correlated significantly with the titers of anti-dsDNA antibody （ r =0. 312, P d0.05） and serum complement C3 level （ r =-0. 315, P 〈0.05）. Conclusion This study suggests that TACI may be involved in the development of LN the level of TACI may be a valuable parameter for evaluating the treatment.
