基质金属蛋白酶-13、白细胞介素-1在大鼠退变腰椎间盘组织中的表达及意义

摘要 目的探讨基质金属蛋白酶-13(MMP-13)、白细胞介素-1(IL-1)在去势大鼠退变腰椎间盘组织中的表达及临床意义。方法 30只3月龄Sprague-Dawley(SD)大鼠,随机均分为3组,每组10只:基础对照组(BL组);手术组(OVX);手术对照组(Sham组);BL组在手术开始前处死,其余2组术后三个月处死,并与处死前10 d和4 d分别给予显色双荧光标记。L2-4椎体进行骨密度及硬组织切片分析,L4-5进行HE常规染色、VG特殊染色及运用免疫组化检测MMP-13、IL-1的表达,观察椎间盘的病理学改变并据评分标准对腰椎间盘的退变程度(LVD)进行评分。结果 1、大鼠腰椎骨量的下降及椎间盘的退化:双侧卵巢切除组比手术对照组严重;2、免疫组化:双侧卵巢切除组与手术对照组比较大鼠椎间盘中MMP-13、IL-1表达明显升高(P<0.05)。结论基质金属蛋白酶-13、白细胞介素-1表达量的增加是腰椎间盘退变形成和发展的重要因素。 Objective To investigate the expression and clinical significance of MMP-13 and IL-1 in degenerated lumbar intervertebral discs in ovariectomized (OVX) rats. Methods Thirty 3-month-old female Sprague-Dawley (SD) rats were divided into 3 groups randomly, the baseline control group (BL Group), the operation group (OVX Group), and the operation control group (Sham Group). Each group had 10 rats. Rats in BL Group were executed before the operation. Rats in the other 2 groups were executed in 3 months after operation. And at the 10th and 4th day before execution, double fluorescence labeling was performed. Bone mineral density (BMD) of the vertebrae 2-4 was tested. Bone histomorphometric analysis was performed. HE staining and VG special staining were performed on the lumbar vertebrae 4-5. The expression of MMP-13 and IL-1 was tested using immunohistochemical method. The pathological changes of intervertebral discs were observed, and the degree of lumbar intervertebral disc degeneration (LVD) was graded using a histological scoring system. Results The decrease of lumbar vertebral bone mass and the degeneration of intervertebral disc in OVX Group were significantly severer than those in Sham Group. The expression of MMP-13 and IL-1 in OVX Group was significantly higher than that in Sham Group (P 〈 0. 05). Conclusion The increase of MMP-13 and IL-1 expression is an important factor in the development and progression of lumbar intervertebral disc degeneration.
