青藤碱抑制肺癌细胞株Calu-1增殖、凋亡影响的实验研究 被引量:8

摘要 目的:探讨青藤碱对肺癌细胞株Calu-1增殖、凋亡影响及相关作用机制。方法:青藤碱150μg/ml、200μg/ml、250μg/ml作用于肺癌细胞株Calu-1,采用MTT、流式细胞术、RT-PCR及Westernblot检测经不同浓度青藤碱处理和空白对照组(0μg/ml),12h、24h、36h后肺癌细胞株Calu-1增殖、凋亡及MEK/ERK蛋白和iASPP基因表达情况。结果:ERK,p-ERK表达随处理浓度的增加而降低,MEK/ERK通路活化受到抑制,在第36h 250μg/ml处理组p-ERK表达最低,且此时iASPP-mRNA基因表达明显降低。不同浓度青藤碱均对肺癌Calu-1细胞株增殖呈现明显抑制并促进其凋亡,随青藤碱浓度增加和作用时间的延长,抑制率增加,促凋亡作用增强,在第36h 250μg/ml处理组抑制率达到(51.23±4.34)%。结论:青藤碱明显抑制肺癌细胞的增殖和凋亡。 Objective:To evaluate the effect of Sinomenine on the proliferation, apoptosis in lung cancer cell lines Calu- 1 and related mechanism. Methods: After deal with different dose of Sinomenine 150μg/ml, 200μg/ml, 250 μg/ml and control group (0 μg/ml) in 12h,24h, 36h later, to use MTT, flow eytometry, RT - PCR and Western - blot to detecte the proliferation, apoptosis, MEK/ERK protein and iASPP gene expression of Calu - 1. Results : MEK/ ERK pathway activation was restrained, ERK 1/2, p - ERK expression decreased with the concentration increased, p - ERK expressed minimum at 36h in the 250μg/ml treatment group ,while the same time iASPP gene expression significant reduced. Calu - 1 was obviously inhibited by Sinomenine, which promoted the apoptosis of Calu - 1 with the concentration increased of Sinomenine. At 3 6 h in 25 0 μg/ml treatment group, the inhibition rate arrived (51.23 _+ 4.34 ) %. Conclusion: Sinomenine significantly inhibited the proliferation and promoted the apoptosis of lung cancer cells.
