摘要 目的研究维甲酸衍生物4-氨基-2-三氟甲基维甲酸酯(ATFMPE)对乳腺癌细胞株MDA-MB-231细胞迁移的影响及可能机制。方法不同浓度维甲酸(ATRA)和ATFMPE处理乳腺癌细胞株MDA-MB-231 48 h后,用MTT法检测ATFMPE对细胞增殖的影响,划痕法检测对细胞迁移的影响,Western blot法检测相关迁移蛋白的表达。结果 AT-FMPE对乳腺癌细胞株MDA-MB-231细胞增殖具有明显的抑制作用,ATFMPE和肌球蛋白轻链激酶(MLCK)抑制剂ML-7对细胞的迁移具有明显的抑制作用,Western blot结果显示ATFMPE显著降低MLCK的表达和肌球蛋白轻链(MLC)的磷酸化。结论 ATFMPE对乳腺癌细胞株MDA-MB-231细胞的迁移具有抑制作用,可能与MLCK表达和MLC磷酸化水平下调相关。 Objective To explore the effect of its probable mechanism of a synthetic retinoid 4-amino-2-tri-fluorom- ethyl-phenyl ester (ATFMPE)on the migration ability of human breast cancer MDA-MB-231 cells. Methods MTT assay was performed to measure the proliferation of MDA-MB-231 cells treated with different concentrations of AT- FMPE. Light microscope was used to observe morphologie changes. The effect of ATFMPE and ML-7, a selective in- hibitor of myosin light chain kinase (MLCK) , on the migration of MDA-MB-231 cells was analyzed by cell migra- tion test. The expression of MLCK and phosphorylation of myosin light chain (MLC) proteins was detected by West- ern blot. Results ATFMPE could inhibit the proliferation of breast cancer MDA-MB-231. ATFMPE and ML-7 could decrease the distances of migration of breast cancer MDA-MB-231 significantly. ATFMPE could inhibit the expression of MLCK and phosphorylation of MLC protein. Conclusion ATFMPE could inhibit the migration of breast cancer MDA-MB-231 cells, which may be related to the down regulation of expression of MLCK and phosphorylation of MLC protein.
