人参皂甙Rg3对肺癌荷瘤小鼠免疫功能影响的研究 被引量:17

摘要 目的探讨人参皂甙Rg3对肺癌荷瘤小鼠免疫功能的影响。方法将48只C57BL/6肺癌荷瘤小鼠随机分为4组:人参皂甙Rg3高剂量组(15 mg/kg)、人参皂甙Rg3低剂量组(5 mg/kg)、顺铂(DDP)组(DDP0.5 mg/kg)和生理盐水组,检测各组给药后荷瘤小鼠的脾自然杀伤细胞(NK)、淋巴因子激活的杀伤细胞(LAK)、细胞毒性T细胞(CTL)活性及血清肿瘤坏死因子(TNF)水平。以正常小鼠12只作为对照组。结果 Rg3低剂量组、Rg3高剂量组、DDP组生存素阳性率明显低于生理盐水组(P<0.05);Rg3低剂量组、Rg3高剂量组瘤体质量小于生理盐水组(P<0.05),但大于DDP组(P<0.01);生理盐水组NK、LAK和CTL活性及血清TNF水平均较正常对照组显著下降(P<0.05);DDP组NK、LAK和CTL活性及血清TNF水平较生理盐水组低(P<0.05);Rg3低剂量组和Rg3高剂量组NK、LAK和CTL活性及血清TNF水平较生理盐水组、DDP组明显提高(P<0.05),但仍低于正常小鼠组(P<0.05);Rg3低剂量组NK、LAK和CTL活性及血清TNF水平与Rg3高剂量组比较,差异无统计学意义(P>0.05)。结论人参皂甙Rg3可明显诱导提高荷瘤小鼠特异性抗肿瘤免疫反应。 Objective To investigate the effect of Ginsenoside Rg3 on the immune function of tumor-bearing mice with lung cancer. Methods Forty-eight tumor-bearing mice with C57BL/6 lung cancer were randomly divided into four groups,high-dose Ginsenoside Rg3 group(Rg3,15 mg/kg), low-dose Ginsenoside Rg3 group(Rg3,5 mg/kg), cisplatin (DDP) group(DDP,0.5 mg/kg) and saline group. The activities of splenic natural killer(NK) cells,lymphokine activated killer(LAK) cells,cytotoxic T lymphocyte(CTL) ,and serum tumor necrosis factor(TNF) level were detected in each group after the administration of tumor-bearing mice was performed. Twelve normal mice as control group. Results The positive rates of Surv-ivin of low-dose Rg3 group,high-dose Rg3 group,DDP group were significantly lower than that of saline group (P 〈0.05) ;Tumor mass of low-dose Rg3 group,high-dose Rg3 group was less than that of saline group (P 〈0.05),but greater than that of DDP group(P 〈0.01 ) ;The activities of NK,LAK and CTL,and serum TNF level decreased significantly in saline group compared with the normal controls(P 〈0.05);The activities of NK,LAK and CTL,and serum TNF level of DDP group were lower than those of saline group ( P 〈 0.05 ), which increased significantly in low-dose Rg3 group and high-dose Rg3 group compared with those of saline group, DDP group ( P 〈 0.05 ), but lower than the normal controls (P 〈 0.05) ;There was no significant difference in the activities of NK, LAK and CTL, and serum TNF level between low-dose P~,3 group and high-dose Rg3 group (P 〉 0. 05 ). Condusion Ginsenoside Rg3 can improve specific anti-tumor immune response in tumor-bearing mice significantly.
