摘要
目的检测血管内皮生长因子C和D(VEGF-C、VEGF-D)及其受体血管内皮生长因子受体3(VEGFR-3)在宫颈癌血清中的表达,并探讨VEGF-C、VEGF-D和VEGFR-3与宫颈癌淋巴转移机制相关性。方法应用酶联免疫分析法(ELISA)检测正常宫颈组10例,宫颈上皮内瘤变CINⅢ组36例,宫颈癌无淋巴结转移患者组33例,宫颈癌有淋巴结转移患者组10例,检测血清中VEGF-C、VEGF-D、VEGFR-3的浓度表达并比较各组间差异性。结果 (1)宫颈癌无淋巴结转移者组VEGF-C、VEGF-D、VEGFR-3血清水平均高于正常宫颈组及CINⅢ组,差异有统计学意义(P<0.05);(2)宫颈癌有淋巴结转移者组VEGF-C、VEGF-D、VEGFR-3血清水平均高于宫颈癌无淋巴结转移组,差异有统计学意义(P<0.05);(3)CINⅢ组VEGF-C、VEGF-D、VEGFR-3血清水平与正常宫颈组相比,差异无统计学无意义(P>0.05)。结论宫颈癌患者血清VEGF-C、VEGF-D和VEGFR-3的表达水平均与淋巴结转移相关,且VEGF-C、VEGF-D与VEGFR-3的血清表达水平密切相关,所以VEGF-C、VEGF-D可能在宫颈癌中通过VEGFR-3信号通道促进淋巴结转移。其可能成为推测宫颈癌淋巴结转移和预后的有用的生物学指标。
Objective To detect vascular endothelial growth factor C and D （VEGF-C and VEGF-D） and their receptor-3 （VEGFR-3） levels in serum of patients with cervical cancer, and to explore the correlation of lymph node metastasis with VEGF-C,VEGF-D and VEGFR-3. Methods ELISA was used to detect the expression levels of VEGF-C ,VEGF-D and VEGFR-3 in serum of 10 cases of normal cervical tissues,36 cases of cervical intraepithelial neoplasia m ~ CIN m group）, 33 cases of cervical cancer without lymph node metastasis, 10 cases of cervical cancer with lymph node metastasis. The difference of expression levels of VEGF-C, VEGF-D and VEGFR-3 were compared among different groups. Results （ 1 ） The expression levels of VEGF-C, VEGF-D and VEGFR-3 in serum of cervical cancer patients without lymph node metastasis were significantly higher than those in normal cervical tissues group and CIN 111 group（ P 〈 0.05 ） ; （ 2 ） The expression levels of VEGF-C, VEGF-D and VEGFR-3 in serum of cervical cancer patients with lymph node metastasis were significantly higher than those in cervical cancer patients without lymph node metastasis（P 〈0.05 ）;（3 ）There was no significant difference in expression levels of serum VEGF-C ,VEGF-D and VEGFR-3 between CIN m group and normal cervical tissues group （ P 〉 0.05 ）. Conclusion The expression levels of VEGF-C,VEGF-D and VEGFR-3 are related to lymph node metastasis in patients with cervical cancer,and the expression levels of VEGF-C and VEGF-D are closely related to that of VEGFR-3. Thus, VEGF-C and VEGF-D may increase the incidence of lymph node metastasis in cervical cancer through VEGFR-3 signaling pathway, which may be used as the valuable indicators for predicting lymph node metastasis and prognosis of cervical cancer.
