摘要
目的采用染色体畸变实验检测艾烟中可吸入颗粒物是否影响细胞DNA结构或改变信息的实现,进而判定艾烟的遗传毒性。方法通过四氮唑盐比色实验得出艾烟冷凝液(MSC)对中国仓鼠卵巢细胞染毒24 h的半数抑制率IC50为0.087 mg/mL,体外培养哺乳动物中国仓鼠卵巢细胞系(CHO),体外代谢活化(S9)及非活化的情况下染毒细胞24 h,随机分为空白组、丝裂霉素C组、1/2 IC50MSC组、1/4 IC50MSC组、1/8 IC50MSC组、空白+S9组、环磷酰胺+S9组、1/2 IC50MSC+S9组、1/4 IC50MSC+S9组、1/8 IC50MSC+S9组。秋水仙胺作用于中期染色体收获细胞,制备染色体标本,观察细胞染色体畸变情况,并进行分析。结果 MSC畸变细胞率和染色体总畸变率由高到低依次为环磷酰胺+S9组>MSC+S9组≥丝裂霉素C组>空白+S9组>MSC组≥空白组。结论艾烟具有生物活性,高浓度的艾烟表现出一定的细胞毒性。
Objective To test the activity of respirable particulate matthers in moxa smoke in vitro genetic toxicology test systems with the incidence of chromosomal aberrations in CHO.Methods For moxibustion smoke condensation(MSC),the IC50 value in Chinese hamster ovary cells harvested 24 h after treatment obtained using MTS/PMS was 0.087 mg/mL.CHO was incubated with S9 mix and harvested 24 h after treatment.Cells were divided into control group,mitomycin C(MMC),1/2 IC50 MSC,1/4 IC50 MSC,1/8 IC50 MSC,control group+S9,cyclophosphamide(CP)+S9,1/2 IC50 MSC+S9,1/4 IC50 MSC+S9,1/8 IC50 MSC+S9.We prepared and analysed metaphase chromosome samples with addition of colchicine.Results The incidence of chromosomal aberrations in CHO,in descending order,were CP+S9,MSC+S9,MMC,control group+S9,MSC,control group.Conclusion MSC has biological activity and showed some cytotoxicity at high-dose.
